HAMSYL® (pegaspargase injection), manufactured by Gennova, is an antineoplastic agent supplied as a 3750 IU / 5 mL single-use vial for intramuscular or intravenous administration. It is indicated as a core component of a multi-agent chemotherapy regimen for treating patients with acute lymphoblastic leukemia (ALL). Pegaspargase works by depleting plasma levels of the amino acid L-asparagine. Unlike normal cells, ALL cells lack asparagine synthetase and rely on external sources; this systemic depletion starves the malignant cells, inducing apoptosis. Treatment requires close clinical monitoring for potential toxicities, including hypersensitivity reactions, pancreatitis, and coagulopathy.
